Mouse Anti-Human Nestin

  • Nestin is a large intermediate filament protein (class Type VI) that is expressed during development, particularly in myotendinous and neuromuscular junctions. Nestin expression is typically restricted to early development stages, disappearing by embryonic day E18. While traditionally considered a neuronal marker, it is also present in endothelial cells, making it a marker for various embryonic tissues and cancerous cells. Nestin is highly expressed in glioblastomas and melanomas but is not present in metastatic carcinoma cells.

    Immunogen: A 12-mer peptide of PEVGDEEALRPL from human Nestin corresponding to amino acids 681–692 (NM_ 006617) was selected as immunogen.

  • Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 μg/ml BSA, and 50% glycerol. Store at –20°C. After receiving, aliquot the antibody and store at –20°C.

  • General notes

    For extended storage, aliquot contents and freeze at -20 °C or below. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4 °C as an undiluted liquid. Dilute only prior to immediate use.

Overview

This monoclonal antibody is specific for Nestin, a type VI intermediate filament protein expressed during development, particularly in myotendinous and neuromuscular junctions. While Nestin is typically absent after embryonic stage E18, it is widely expressed in several cell types including endothelial cells. Notably, Nestin is used as a marker for neural progenitor cells and is prominently found in glioblastoma multiforme (GBM) and melanomas (both primary and metastatic), making it valuable in cancer research. It is also useful in identifying various embryonic tissues, although its expression is not exclusive to the CNS.

  • Example Applications

    1. Indirect intracellular staining of bovine Sertoli cells and flow cytometry analysis using anti-nestin (4G10G8) antibody: FITC-conjugated rabbit anti-mouse was used as a secondary antibody.
    2. Western blot assay using affinity-purified anti-nestin (4G10G8): Cell lysates from several cell lines were analyzed using 6% SDS-PAGE under reducing and non-reducing conditions, revealing several bands between 150–300 kDa.
    3. Immunocytochemical analysis of nestin expression in purified bovine Sertoli cells: Immunostaining revealed co-expression of nestin and vimentin, with U373 cell line as the positive control.
